Comments: This line is IL-3 dependent.
The line was originally contaminated with mycoplasma, and was cured by treatment with BM-cycline.
The cells were assayed for mycoplasma, by the Hoechst stain, PCR and the standard culture test, after a six-week period following treatment. All tests were negative.
Propagation: 32D Clone 3细胞ATCC complete growth medium: RPMI 1640 medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES, and 1.0 mM sodium pyruvate supplemented with:10% heat-inactivated fetal bovine serum10% mouse Interleukin-3 culture supplement (available from Becton Dickinson Cat. No. 354040, formerly Collaborative Research Cat. No. 40040)

Subculturing: Protocol: Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 X 10 exp5 viable cells/ml. Maintain cell density between 2 X 10 exp5 and 1 X 10 exp6 viable cells/ml. 32D Clone 3细胞Scrape off the attached cells and transfer along with the floating cells into new flasks.
