Cytogenetic Analysis: modal number = 66; range = 51 to 90
The modal number (66) does not permit sexing of donor cytologically. Most cells contain one long, unpaired, submetacentric chromosome and 2 to 3 small telocentric chromosomes with satellites. Polyploidy was observed in 5/200 metaphase cells.
Comments: The SIRC cell line was derived by M. Volkert of the Staatens Seruminstitut, Copenhagen, Denmark, from the cornea of a normal rabbit in 1957. Little is recorded relating to the history of this cell line for approximately the first 400 passages. In 1965, J. Leerhoy [PubMed ID: 14331182] found this line to be susceptible to rubella virus. C.A. Phillips, et al. [PubMed ID: 5918951] confirmed the above findings and demonstrated the suitability of the cell line for primary isolation of rubella virus. The early appearance of distinct cytopathic changes makes this cell line highly suitable for both the propagation and quantitation of rubella virus [PubMed ID: 6042441].
Propagation: SIRC细胞ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Eagle's Minimum Essential Medium, Catalog No. 30-2003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.

Subculturing: Protocol:
Remove and discard culture medium.
Briefly rinse the cell layer with 0.25% (w/v) Trypsin - 0.53 mM EDTA solution to remove all traces of serum which contains trypsin inhibitor.
Add 2.0 to 3.0 ml of Trypsin-EDTA solution to flask and observe cells under an inverted microscope until cell layer is dispersed (usually within 5 to 15 minutes).
Note: To avoid clumping do not agitate the cells by hitting or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach may be placed at 37C to facilitate dispersal.
Add 6.0 to 8.0 ml of complete growth medium and aspirate cells by gently pipetting.
Add appropriate aliquots of the cell suspension to new culture vessels.
Incubate cultures at 37C.
Subcultivation Ratio: A subcultivation ratio of 1:4 to 1:8 is recommended
