Comments: TK-1细胞TK1 is a T-cell lymphoma cell line originated by E. C. Butcher and I. L. Weissman from a spontaneous thymic lymphoma/leukemia that developed in an AKR/Cum mouse. [38634]
TK1 cells have a unique phenotype with high levels of the lymphocyte integrin, alpha 4, beta 7.
They bind constitutively to the murine mucosal vascular addressin, MAdCAM-1. [22636]
TK1 cells are useful for studies of alpha 4, beta 7 function and regulation. The cells are CD8 positive; they display approximately 1% of L-selectin of that expressed by normal lymph node cells.
Propagation: ATCC complete growth medium: RPMI 1640 medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES, and 1.0 mM sodium pyruvate and supplemented with 0.1 mM non-essential amino acids, 0.05 mM 2-mercaptoethanol, 90%; fetal bovine serum, 10%

Subculturing: Protocol: Cultures can be maintained by addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension in fresh medium at 1 to 2 X 10(5) viable cells/ml. Do not allow the cell concentration to exceed 2 X 10(6) cells/ml.
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)
