Comments: Animals were immunized with the ASPC-1 human pancreatic carcinoma cell line.
Spleen cells were fused with NS-1 myeloma cells.
The antibody reacts with some colon and pancreatic carcinoma cell lines as well as some T cell leukemia lines.
AS 33细胞In tissues, the reactivity is restricted to carcinomas and normal colonic mucosa.
Some reactivity was also detected in salivary gland and breast tissue.
Other normal tissues test were negative.
The AS 33 antigen is heat stable.
Propagation: ATCC complete growth medium: RPMI 1640 medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES, and 1.0 mM sodium pyruvate and supplemented with 0.1 mM hypoxanthine and 0.016 mM thymidine, 90%; fetal bovine serum, 10%

Growth Conditions: These cells must be grown using a feeder layer. Mouse peritoneal exudate cells or irradiated mouse fibroblasts (e.g., irradiated STO cells, ATCC 56-X) will provide suitable feeder layers.
Subculturing: Protocol: Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 2 X 10(5) cells/ml and maintain between 1 X 10(5) and 1 X 10(6) cells/ml.
Medium Renewal: Every 2 to 3 days
