Cytogenetic Analysis: This is a hypertetraploid human cell line with a modal number between 93 and 99. The frequency of cells with higher ploidies is 2.0%. However, since the stemline chromosome number is high, the estimate for the polyploidy is tentative. Thirteen or more marker chromosomes were common to all cells. Daoy细胞Of these, many had two to four copies per cell. Among the markers were: t(1q5q), t(13q;?), 15p+, 7q+, der(9)t(3;9)(p21;q34) and eight others. In most cells, the 15p+ has three copies and der(9) has four copies.Some cells have del(1)(p11). Normal N12, N14, N15 and N19 tend to have four or more copies per cell. There are two normal X chromosomes in most cells, but there is no detectable normal Y.

Comments: Daoy细胞The Daoy cell line was established in 1985 by P. F Jacobsen of the Royal Perth Hospital in Western Australia.
The line was derived from biopsy material taken from a tumor in the posterior fossa of a 4 year old boy.
Although the original tumor had characteristics of both neuronal and glial differentiation, these were not retained by the cell line.
Treatment of the cells with dibutyryl cyclic amp (cAMP) does not induce expression of those characteristics as measured by staining for S100 (S-100) protein and glial fibrillary acidic proteins (GFAP).
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Eagle's Minimum Essential Medium, Catalog No. 30-2003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.

Subculturing: Protocol:
Remove and discard culture medium.
Briefly rinse the cell layer with 0.25% (w/v) Trypsin- 0.53 mM EDTA solution to remove all traces of serum that contains trypsin inhibitor.
Add 2.0 to 3.0 ml of Trypsin-EDTA solution to flask and observe cells under an inverted microscope until cell layer is dispersed (usually within 5 to 15 minutes).
Note: Daoy细胞To avoid clumping do not agitate the cells by hitting or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach may be placed at 37�C to facilitate dispersal.
Add 6.0 to 8.0 ml of complete growth medium and aspirate cells by gently pipetting.
Add appropriate aliquots of the cell suspension to new culture vessels.
Incubate cultures at 37�C.
Subcultivation Ratio: A subcultivation ratio of 1:4 to 1:6 is recommended
