Comments: This line was cloned by limiting dilution from the A375 melanoma cell line (see ATCC CRL-1619).
These cells will eventually die and lyse when cultured in medium containing interleukin-1 (interleukin 1, IL-1) and therefore can be used in a reliable and sensitive growth inhibition assay for IL-1.
Sensitivity to IL-1 is maximal when the cells are derived from subconfluent cultures in logarithmic growth.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Eagle's Minimum Essential Medium, Catalog No. 30-2003. A375.S2细胞To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.

Subculturing: Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:8 is recommended
Medium Renewal: Every 2 to 3 days
Remove medium, rinse with fresh 0.25% trypsin, 0.03% EDTA solution and let the culture sit at room temperature (or 37C) until cells detach (about 10 minutes). Add fresh medium, aspirate and dispense into new flasks.
Preservation: culture medium 95%; DMSO, 5%
