储 存 液0.01M TBS(pH7.4) with 1% BSA, 0.03% Proclin300 and 50% Glycerol.
保存条件Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
Histone H3.1抗体特异性问题。从本质上来说,抗体的工作原理就是与特定蛋白或其他抗原结合,从而实现定性,定量,定位等检测。所以,抗体能否**的结合抗原,是成败的关键,抗体的特异性(specificity)就显得尤其重要了。而对特异性影响*大的,莫过于抗体的交叉反应性(cross-reactivity)。在特异性正常情况下,Histone H3.1抗体只能结合**的一种蛋白。而如果抗体本身出现问题(如结构等),可能会结合其它非目标蛋白,从而导致特异性降低。更令人恼火的是,如果抗体的特异性问题很难被检测出来。假设某个糟糕的抗体X理论上只结合蛋白A,但是它也可以结合蛋白B。当实验样本里蛋白B没有出现时,它将完全得到完全正确的实验结果。如果当样本中包含蛋白B,偏差就出现了。试想一下,你在应用抗体X时,如果你在项目前期没有使用过蛋白B样本,一切都会很顺利。但是在项目后期,你开始加入蛋白B样本,你的实验结果就完全乱了套。那么问题就来了:你事先并不知道抗体X可以结合非目标蛋白B,所以你完全无法解释为什么项目的前后会出现偏差。
Histone H3.1抗体出现交叉反应,导致特异性降低,主要是由抗体本身的质量问题引起的。由于抗体本身结构和生产过程的复杂性,生产商稍有疏忽,抗体质量就会出现问题。由于抗体是科研的刚需产品,利润又很高,所以引得众多公司疯狂进入到这个市场,直接导致了市面抗体产品质量良莠不齐,供应商鱼龙混杂。而市场本身又缺少有力的监督和管理机制,抗体频频出现质量问题也就不足为奇了。
