Description | The Immortalized Mouse OP9 Stromal Cells (I-OP9) is a reliable source of M-CSF-deficient mouse stromal cells which has been reported to be able to support the differentiation of pluripotent stem cells (iPSCs), such as hematopoietic stem cells (HSC). The OP9 cells were immortalized through viral infection with retrovirus carrying the LXSN16E6E7 vector to generate I-OP9. I-OP9 cells are tumorigenic when injected into mice. However, treatment with mitomycin C before in vitro differentiation will eliminate the tumorigenicity issue before using as feeder cells for HSC. Additionally, the Immortalized Mouse OP9 Stromal Cells (I-OP9) may be used to study functions of M-CSF deficiency and osteroporosis. |

Propagation | Use of PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels with the following conditions. The base medium for this cell line is PriGrow VIII medium available from ABM (TM018). To make the completed growth medium, add the following components to the base medium: fetal bovine serum (TM999) to final concentration of 20%, 1X non-essential amino acids, 2mM L-glutamine (G275), 1 mg/mL G418, and Penicillin/Streptomycin (G255). Atmosphere: air: 95%, CO₂: 5%; Temperature: 37.0°C. |
Quality Control | 1) Cells are tested to propagate over 42 passages, and retained phenotypes similar to the primary cells; 2) HPV gene was detected via RT-PCR. |
