Membrane. Cell junction > synapse > postsynaptic cell membrane. Enriched in post-synaptic plasma membrane and postsynaptic densities (PSD). Accumulates in spines along with synapse maturation and colocalizes with DLG4 in a punctate pattern.
一抗操作步骤:
洗涤:洗涤的目的是为了洗去一抗的非特异性结合,洗涤的效果直接影响结果背景的深浅。建议使用PBST或TBST作为洗涤液,其中的Tween20有助于去除非特异性的结合,减少背景。洗涤的次数和时间根据具体实验来确定,建议清洗3-5次,每次5分钟。
