标准液配制方法：

取400NTU的标准原液50mL注入容量瓶内，再取950mL纯水注入容量瓶内、摇晃混匀，此液体为20NTU标准液。

二、10NTU 标准液配制方法：

取400NTU的标准原液25mL注入容量瓶内，再取975mL纯水注入容量瓶内、摇晃混匀，此液体为10NTU标准液。

三、配制的10NTU及20NTU标准液，要现配现用，常温下存放，就会变化，不能使用。

四、400NTU 标准液，不能在常温下存放，浊度标准液应贮存于1—10℃以内环境中(放置冰箱冷藏室内贮存)。

标准溶液配制：

1。先將PH7.00同PH4.00藥粉分別倒入250毫升蒸餾水中，充分溶解，用兩支玻璃瓶密封保存，做好標簽。

2 。在校正前，準備ORP標準溶液。

3 。86毫伏：取50至100毫升PH7.00標準溶液置入一玻璃杯中，再加入稍許醌氫醌，充分攪拌。

4 。256毫伏：取50至100毫升PH4.00標準溶液置入一玻璃杯中，再加入稍許醌氫醌，充分攪拌。
5。 ORP標準液保存期為72hour。

注意事项：

1.流动相必须用HPLC级的试剂，使用前过滤除去其中的颗粒性杂质和其他物质(使用0.45um或更细的膜过滤)。

2.流动相过滤后要用超声波脱气，脱气后应该恢复到室温后使用。

3.不能用纯乙腈作为流动相，这样会使单向阀粘住而导致泵不进液。

4.使用缓冲溶液时，做完样品后应立即用去离子水冲洗管路及柱子一小时，然后用甲醇(或甲醇水溶液)冲洗40分钟以上，以充分洗去离子。对于柱塞杆外部，做完样品后也必须用去离子水冲洗20ml以上。

5.长时间不用仪器，应该将柱子取下用堵头封好保存，注意不能用纯水保存柱子，而应该用有机相(如甲醇等)，因为纯水易长霉。

6.每次做完样品后应该用溶解样品的溶剂清洗进样器。

7.C18柱不能进蛋白样品，血样、生物样品。

8.堵塞导致压力太大，按预柱→混合器中的过滤器→管路过滤器→单向阀检查并清洗，清洗方法；①以异丙醇作溶剂冲洗：②放在异丙醇中间用超声波清洗；

9.气泡会致使压力不稳，重现性差，所以在使用过程中要尽量避免产生气泡。

10.如果进液管内不进液体时，要使用注射器吸液：通常在输液前要进行流动相的清。

11.要注意柱子的PH值范围，不得注射强酸强碱的样品，特别是碱性样品。

12.更换流动相时应该先将吸滤头部分放入烧杯中边振动边靖洗，然后插入新的流动相中。

对照品实验方法与判定：

1.对照品溶液的稳定性

2.对照品溶液的配制：精密量取脑对照品适量，加无水乙醇制成每1ml含脑1mg的溶液，作为对照品溶液。

3.色谱条件：以交联键合聚乙二醇为固定相的毛细管柱；柱温120℃；进样口温度250℃；检测器温度250℃；分流进样，分流比10:1。理论塔板数按脑计算应不低于10000。

4.实验方法：配制的对照品溶液，一份置冷处保存，一份置室温中保存，在第1、3、7、10、15天重新配制一份对照品溶液，三种储备液同时稀释制成每1ml中约含50ug的溶液。照气相色谱法，并依据新对照品的峰面积计算原对照品溶液的含量。记录下来，保证原对照品溶液含量在考察期相对平均偏差不超过2.0%，验证对照品溶液在使用期内稳定可靠。
