Goodjob 操作流程
1:具备传统的照胶功能,可用于化学放光分析,多色荧光,实时电泳等。
2:PCR(polymerase chain reaction):聚合酶链式反应。
(1)DNA变性(90-96℃):热作用,氢键断裂为单链DNA.
(2)退火(25-65℃):引物与DNA模板结合,形成局部双链.
(3)延伸(70-75℃):Taq酶(DNA聚合酶)作用下,以dNTP为原料,从引物3-5延伸,合成与模板互补DNA.

(5) Turn on main power、CCD、white light、mini-pc
打开主电源、CCD、白光灯、迷你电脑
(6) Auto-booting to capture window 自动启动捕捉窗口
(7) Start to load sample active Loop icon to see realtime image 开始加载样品,选择Loop观察实时图像
Adjust: zoom,focus aperture always set to maximum.routine use don’t need to adjust,only do it when size change or out of focus.
校准:变焦,光圈通常设置为*大值。日常使用不需要校准,仅当样品规格改变或超出聚焦范围时校准。
(8) adjust zoom/focus 调整变焦/焦距
(9) start to capture time-lapse samples 开始延时捕捉样品图像
turn off loop icon 关闭Loop
click time-lapse icon 点击Time-lapse
(10)change capture image no. 改变捕捉图像数量
(11)set exposure time. bin no. auto-save image. Next dir 设置曝光时间、bin值、自动保存图像、下一目录
Pre-set 预先设置
1)auto-stretch for non-saturate bands auto-stretch用于浅色条带
2)negative for dark bands image and white background negative 用于深色条带和白色背景
(12)turn off reflectant white light.press start 关闭反射白光,按开始
1)the first image to last image maintain same intensity,this lead to high concentration bands not easy to saturated. **到*后一张图像保持相同的强度,这使得高强度的条带不易被饱和。
2)turn off auto-stretch,this lead to image more like X-ray film and easy to see high concentration bands over exposure. 关闭auto-stretch,这会使图像更像X光片,并且很容易发现高强度的条带曝光过度。
(13)choose any image after capture finish. 捕捉完后选择任一图像
Time-lapse capture
Expose=0.5sec./per fame
Set 0-10 images or more
Bin=1,auto-stretch.
(14) turn off white light image capturing
Time-lapse capture
Expose=1sec./per fame
Set 0-10 images or more
Bin=1,auto-stretch.
