收到人羊膜间充质基质细胞时如无异常情况，请在显微镜下观察细胞密度，如为贴壁细胞，未超过80%汇合度时，将培养瓶中培养液吸出，留下10ml培养液继续培养；超过80%汇合度时，请按细胞培养条件传代培养。如为悬浮细胞，吸出培养液、1000转/分钟离心2分钟，吸出上清，管底细胞用新鲜培养基悬浮细胞后移回培养瓶。

Multipotent mesenchymal stramal cells, also known as mesenchymal stem cells (MSC) are well-characterized population of ***** stem cells. They have the potential to develop into mature cells that produce fat, cartilage, bone, tendons, and muscle [1]. These properties in combination with their developmental plasticity have generated tremendous interest in the potential use of mesenchymal stem cells to replace damaged tissues. MSC cultured without serum in the presence of transformation growth factor will differentiate into chondrocytes, whereas MSC cultured in serum with ascorbic acid and dexamethasone will differentiate into osteoblasts. MSC has the capability for renewal and differentiation into various lineages of mesenchymal tissues. MSC derived from fetal membrane such as amnion and chorion express CD271 [2], possess characteristics of cardiomyocytes [3], and various antiagiogenic and anti-inflammatory proteins.

HAMSC from ScienCell Research Laboratories are isolated from human amniotic membrane. HAMSC are cryopreserved at passage one culture and delivered frozen. Each vial contains >5 x 10^5 cells in 1 ml volume. HAMSC are characterized by immunofluorescent method with antibodies to CD73, CD90, CD105 and lipid staining after differentiation. HAMSC are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. HAMSC are guaranteed to further culture at the conditions provided by ScienCell Research Laboratories.

Recommended Medium

It is recommended to use Mesenchymal Stem Cell Medium (MSCM, Cat. No. 7501) for the culturing of HAMSC in vitro.

Storage

Directly and immediately transfer cells from dry ice to liquid nitrogen upon receiving and keep the cells in liquid nitrogen until cell culture is needed for experiments.
