Applications:	The TC-1 lung metastasis model can be used to test the efficacy of various E6/E7-specific vaccines and immunotherapeutic strategies [PubMed: 9724092]. The tumor cell line, TC-1 [tissue culture number one], was derived from primary lung epithelial cells of C57BL/6 mice. The cells are positive for the expression of HPV-16 E7 [PubMed: 8548765].

Comments:	The tumor cell line, TC-1 [tissue culture number one], was derived from primary lung epithelial cells of C57BL/6 mice. The cells were immortalized with the amphotropic retrovirus vector LXSN16E6E7 and subsequently transformed with the pVEJB plasmid expressing the activated human c-Ha-ras oncogene [PubMed: 8548765]. The transformed cells were selected with G418 and Hygromycin B [PubMed: 8548765]. The cells are positive for the expression of HPV-16 E7 [PubMed: 8548765].The TC-1 lung metastasis model can be used to test the efficacy of various E6/E7-specific vaccines and immunotherapeutic strategies [PubMed: 9724092].
Propagation:	ATCC complete growth medium: RPMI 1640 medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES, 1.0 mM sodium pyruvate supplemented with 0.1 mM nonessential amino acids, 90%; fetal bovine serum, 10% Atmosphere: air, 95%; carbon dioxide (CO2), 5% Temperature: 37.0°C
Subculturing:	Protocol: Subculture before confluence. Remove and discard culture medium. Briefly rinse the cell layer with 0.25% (w/v) Trypsin- 0.53 mM EDTA solution to remove all traces of serum that contains trypsin inhibitor. Add 2.0 to 3.0 ml of Trypsin-EDTA solution to flask and observe cells under an inverted microscope until cell layer is dispersed (usually within 5 to 15 minutes). Note: To avoid clumping do not agitate the cells by hitting or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach may be placed at 37°C to facilitate dispersal. Add 6.0 to 8.0 ml of complete growth medium and aspirate cells by gently pipetting. Add appropriate aliquots of the cell suspension to new culture vessels. Cultures can be established between 1 X 10(5) and 2 X 10(5) viable cells/cm2.Maintain cultures at a cell concentration between 2 X 10(5) and 4 X 10(5) viable cells/cm2. Do not exceed 6 X 10(5) cells/cm2.. Incubate cultures at 37°C. Interval: A subcultivation ratio of 1:3 to 1:10 is recommended Medium Renewal: Every 2 to 3 days
