Description | The Aryl hydrocarbon receptor (AhR) is a ligand-activated transcriptional factor that regulates genes implicated in drug metabolism, cell proliferation, immune responses, circadian rhythm and tumor promotion. Due to the numerous and varied effects that AhR activation may have on the cell, research investigating novel AhR ligands is important for drug interactions and chemically induced carcinogenesis. The AZ-AHR Stable HepG2 Luciferase Reporter Cell Line is a sophisticated and highly sensitive tool for gene reporter assays through the dose- and time-dependent induction of luciferase activity in response to various AhR ligands. This immortalized cell line also remains fully functional for over 15 passages increasing efficiency and consistency between experiments. |

Recommended Seeding Density | Thaw entire contents into an appropriate T25 flask as specified in the Propagation instructions. |

Propagation | Use of PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels with the following conditions. The base medium for this cell line is Prigrow III medium available in ABM, Cat. No. TM003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999) to final concentration of 10%, 4 mM L-glutamine, 1% nonessential amino acids, 1mM sodium pyruvate and Penicillin/Streptomycin (G255). Atmosphere: air: 95%, CO₂: 5%; Temperature: 37.0°C. |
